Wayne Elliot Knight was born on August 7, 1955, in New York City to Grace (née Monti) and William Knight. His family relocated to Cartersville, Georgia, where his father worked in the textile industry. Raised Catholic, Knight played on his high school football team as a lineman. He attended the University of Georgia before pursuing an acting career, eventually returning to complete his degree in 2008.

Knight's professional training began with an internship at the Barter Theatre in Abingdon, Virginia, a repertory company through which he earned his Equity card. He then returned to New York City, and after two years landed his first Broadway role. His stage career spanned from 1977 to 2012 and included productions of Gemini, Mastergate, Sweet Charity, Art, and Elf. He appeared in Art in 1999 and returned to Broadway in November 2012 to play Santa in Elf: The Musical. His Sweet Charity credit featured Christina Applegate. Between his stage engagements, Knight also worked for five years as a private investigator.

His film career began in the late 1980s with appearances in Dirty Dancing (1987) and Born on the Fourth of July (1989), followed by Dead Again (1991), JFK (1991), and Basic Instinct (1992). Director Steven Spielberg, having seen Knight's work in Basic Instinct, cast him as the first actor in Jurassic Park (1993). Knight played Dennis Nedry, InGen Corporation's chief programmer and secret spy for the rival company Biosyn, a role that earned him a Saturn Award for Best Supporting Actor nomination. He went on to appear in To Die For (1995), Space Jam (1996), For Richer or Poorer (1997), Rat Race (2001), and Punisher: War Zone (2008), in which he played the techno sidekick Microchip. A romantic comedy, She Wants Me, featured Knight in April 2012, and he is set to appear in Five Nights at Freddy's 2 (2025).

On television, Knight became widely recognized for two recurring roles on NBC. From 1992 to 1998 he played Newman, Jerry Seinfeld's neighbor and nemesis, on Seinfeld, and from 1996 to 2001 he portrayed Officer Don Orville, Sally's love interest, on 3rd Rock from the Sun. He reprised the Newman character in the seventh season of Curb Your Enthusiasm in November 2009 and again in a Super Bowl commercial in February 2014 alongside Jerry Seinfeld and Jason Alexander. Knight also appeared on sketch comedy series The Edge for Fox and Assaulted Nuts for Cinemax and Channel 4 in the United Kingdom, and won the blue door award on the American version of Thank God You're Here. Additional television credits include guest appearances on Bones, CSI: Crime Scene Investigation, The Whole Truth, and Hot in Cleveland, where he became a recurring character in the second season. He played Brian Friedkin in the BBC/Starz series Torchwood: Miracle Day in the summer of 2011 and appeared as a regular on the TV Land sitcom The Exes.

Knight has contributed extensively to animated productions. He voiced Tantor the elephant in Tarzan (1999) and Al McWhiggin, the manager of Al's Toy Barn, in Toy Story 2 (1999). His television voice roles include Igor on Toonsylvania (1998–1999), Dojo Kanojo Cho in Xiaolin Showdown (2003–2006), Mr. Blik on Catscratch (2005–2007), Emperor Zurg on Buzz Lightyear of Star Command, Demetrius in Hercules (1997), and Baron Von Sheldgoose on Legend of the Three Caballeros (2018). He also voiced the Elf Elder in Tom and Jerry: The Lost Dragon (2014) and made guest voice appearances on Billy & Mandy's Jacked-Up Halloween, Dilbert, Brandy & Mr. Whiskers, and The Penguins of Madagascar.

Knight married makeup artist Paula Sutor on May 26, 1996, in a ceremony held at the home of Seinfeld cast member Michael Richards; the couple divorced by December 2003. He married his second wife, Clare de Chenu, on October 15, 2006, and they have a son named Liam. The family resides in Toluca Lake, Los Angeles.
