Thomas Francis Rosqui was born on June 12, 1928, in Oakland, California, to Sally Rosqui, a homemaker of Portuguese descent, and Thomas A. Rosqui, an Italian-American traffic manager. He grew up in Sacramento, where he attended Sacramento Junior College before earning a drama scholarship to the College of the Pacific. There he participated in college productions and received the school's F. Melvyn Lawson Award in dramatics.

Rosqui's professional acting career began in 1951 with an appearance at St. John Terrell's Music Circus in Sacramento, in a production of Show Boat. He subsequently joined the national touring company of The Cocktail Party. During his service in the U.S. Navy, he performed with the Actor's Workshop in San Francisco. Following his discharge, he relocated to New York, though he returned to San Francisco to appear in Gift of Fury, a play by Herbert Blau, who co-led the San Francisco Actors' Workshop alongside Jules Irving. When Blau and Irving established the Lincoln Center Repertory Company, Rosqui was invited to join the company, spending two years there and appearing in productions including The Condemned of Altona and Danton's Death.

His Broadway career spanned from 1965 to 1973 and included productions such as Yerma, The Alchemist, East Wind, Epstein, and The Lincoln Mask, as well as Sticks and Bones and The Price. Beyond the stage, Rosqui became widely recognized for his film work, most notably as Rocco Lampone, a bodyguard to the Corleone family, in the first two Godfather films. He also made television appearances throughout his career, and by the 1980s had returned primarily to stage work.

Rosqui was married to actress Erica Yohn. He died on April 12, 1991, at the age of 62, from cancer, and was survived by a son and two stepchildren.
