Nnamdi Asomugha was born on July 6, 1981, in Lafayette, Louisiana, to Nigerian Igbo parents. He was raised in Los Angeles, California, attending Leuzinger High School in Lawndale, Bishop Montgomery High School in Torrance, and Narbonne High School in Harbor City, where he played both basketball and football. He went on to study at the University of California, Berkeley, competing for the California Golden Bears football team and finishing his collegiate career with 187 tackles, eight interceptions, three touchdowns, and 15 pass deflections across 41 games. He graduated with a Bachelor of Science in Business Administration.

The Oakland Raiders selected Asomugha with the 31st overall pick in the first round of the 2003 NFL Draft, making him the second Cal player taken in the first round that year. He spent eight seasons with the Raiders, initially playing sparingly before becoming a starter in 2005. His 2006 season proved a turning point: he recorded eight interceptions, tying for the second-highest total in the league that year, and earned a spot on both the Dr. Z Sports Illustrated and Associated Press All-Pro teams. The Raiders named him their Most Valuable Player and presented him with the Commitment to Excellence Award. He was named team captain for the 2007 season and was among the least-targeted cornerbacks in the league, with opposing quarterbacks attempting only 31 passes in his direction that year, resulting in just 10 completions. In February 2008, the Raiders placed the exclusive franchise tag on him, and he was later selected as a starter for the 2009 Pro Bowl. That same offseason, the Raiders re-signed him to a three-year deal that made him the highest-paid defensive back in NFL history at the time, with the first two years worth $28.5 million in fully guaranteed money.

Asomugha was voted All-Pro four times during his career, including twice to the first team, and was selected to the Pro Bowl three times while with Oakland. Fox Sports and USA Today both named him to their NFL All-Decade Team for the 2000s, and the Oakland Tribune identified him as one of the greatest Raiders of all time. After his tenure with Oakland concluded, he played for the Philadelphia Eagles and the San Francisco 49ers before retiring following the 2013–2014 season, completing an 11-year NFL career.

Following his retirement from professional football, Asomugha pursued careers in acting and producing. He received critical recognition for his portrayal of Carl King in the 2017 film Crown Heights, earning an Independent Spirit Award nomination for Best Supporting Male and an NAACP Image Award nomination for Outstanding Supporting Actor in a Motion Picture. Variety named him one of seven breakout performers of 2017. In 2020, he produced and starred in Sylvie's Love, which received a Primetime Emmy Award nomination for Outstanding Television Movie — his first Emmy nomination as a producer. That same year, Variety included him on its list of ten producers to watch.

Asomugha made his Broadway debut in 2020 in the Pulitzer Prize-winning play A Soldier's Play, portraying Private First Class Melvin Peterson, a role first performed off-Broadway by Denzel Washington. The production won the Tony Award for Best Revival of a Play. Asomugha subsequently received a Tony Award nomination for Best Revival of a Play in 2024, along with a Drama Desk Award nomination for Outstanding Revival of a Play the same year.
