She is married to actor Justin Hagan, and the couple have two daughters, Ruby and Iris, born in 2009 and 2013 respectively.

After relocating to New York City, Shor built her early stage career Off-Broadway, most notably originating a role in the rock musical Hedwig and the Angry Inch, in which she performed from 1998 to 2000. Her Broadway career includes an appearance in 1994 and credits in Fiddler on the Roof. In 2015, she portrayed Madelaine True in the Encores! production of The Wild Party at New York City Center, appearing alongside her Younger co-star Sutton Foster.

Shor reprised her role of Yitzhak in the 2001 film adaptation of Hedwig and the Angry Inch, having previously taken a supporting part in the Harold Ramis comedy Bedazzled in 2000. Her other film work includes The Cake Eaters (2007), in which she supported Mary Stuart Masterson's directorial debut, and Shortbus (2006), directed by Hedwig co-star John Cameron Mitchell. From 2015 to 2021, Shor starred as Diana Trout in the TV Land and Paramount+ comedy-drama series Younger.
