Marie Mireille Enos was born on September 22, 1975, in Kansas City, Missouri, to Jon Goree Enos, an American from Texas, and Monique Enos, a French teacher originally from France. Enos speaks French fluently. She has two brothers and two sisters, including an older sister, Veronique Enos Kaefer, who serves as vice president of philanthropy at The Michael J. Fox Foundation. The family relocated to Sugar Land, Texas, when Enos was five years old. She received her early acting training at Houston's High School for the Performing and Visual Arts, then went on to study theater at Brigham Young University. During her time at BYU, she won the Irene Ryan Award at the Kennedy Center in Washington, D.C., a prize given annually to the nation's top collegiate actor. She left BYU before completing her third year to pursue an acting career in New York City.

Enos made her screen debut in the 1994 television film Without Consent and her feature film debut in the 2001 romantic comedy Someone Like You, starring Ashley Judd. Her Broadway career spanned from 2001 to 2005 and included appearances in The Invention of Love, Absurd Person Singular, and Who's Afraid of Virginia Woolf? Her performance in the 2005 Broadway revival of Who's Afraid of Virginia Woolf?, which starred Kathleen Turner and Bill Irwin, earned her a Tony Award nomination for Best Featured Actress in a Play. During this period she also appeared regionally, playing Perdita in the Shakespeare Theatre Company's 2002 production of The Winter's Tale at the Lansburgh Theater in Washington, D.C.

In 2007, Enos joined the cast of the HBO drama series Big Love, portraying twin sisters JoDean Marquart and Kathy Marquart as a regular cast member in the third and fourth seasons. Her television career expanded significantly in 2010 when she was cast in the lead role of homicide detective Sarah Linden in the AMC drama series The Killing, based on the Danish series Forbrydelsen. The series ran for four seasons, concluding in 2014. For that role, Enos received a Primetime Emmy Award nomination for Outstanding Lead Actress in a Drama Series, a Golden Globe Award nomination for Best Actress in a Television Series Drama, and three Saturn Award nominations for Best Actress on Television.

Following her work on The Killing, Enos appeared in several feature films. In 2013 she played Karin Lane, the wife of Brad Pitt's character, in World War Z, and co-starred as Josh Brolin's character's wife in Gangster Squad. That same year she appeared opposite Reese Witherspoon in Devil's Knot, a crime drama based on the true story of the West Memphis Three. In 2014 she appeared in the crime thriller Sabotage, directed by David Ayer, and starred in If I Stay, directed by R. J. Cutler and based on the novel by Gayle Forman. Also that year she co-starred alongside Ryan Reynolds in The Captive, directed by Atom Egoyan. She later took the lead role in the thriller Never Here, directed by Camille Thoman, and starred in the independent film Katie Says Goodbye alongside Olivia Cooke, as well as Behold My Heart, directed by Joshua Leonard.

In March 2015, Enos was cast as the lead character Alice Vaughan in the ABC legal drama series The Catch, produced by Shonda Rhimes. The show, in which Enos played the head of a high-end private detective agency, aired a ten-episode first season in spring 2016 and a second ten-episode season in 2017 as part of ABC's Thursday night lineup. In 2019, she appeared as Marissa Wiegler in the Amazon Prime Video action drama series Hanna, a role originated by Cate Blanchett in the 2011 film of the same name. Her 2018 film The Lie, a reunion with The Killing showrunner Veena Sud, was released on October 6, 2020, as part of Amazon Prime Video's Welcome to the Blumhouse horror anthology.

Enos married actor Alan Ruck on January 4, 2008. They have two children together, and she has two stepchildren from Ruck's previous marriage. She was raised in the Church of Jesus Christ of Latter-day Saints but is no longer a practicing member.
