Jeffrey Matthew Settle was born on September 17, 1969, in Hickory, North Carolina, the youngest of six children born to Robert Settle, a Baptist minister, and Joan Settle. He has two sisters and three brothers.

Settle's Broadway activity spans from 1996 to 2008 and includes credits in The 24 Hour Plays 2004 and The Yellow Brick Road Not Taken. He stepped into the role of Billy Flynn in Chicago: The Musical for a strictly limited engagement running from April 19 through June 13, 2010.

On screen, Settle first gained attention through his feature film debut in I Still Know What You Did Last Summer, in which he played Will Benson alongside Jennifer Love Hewitt, Freddie Prinze Jr., Mekhi Phifer, and Brandy Norwood. He went on to portray Captain Ronald Speirs in the 2001 HBO miniseries Band of Brothers and Jacob Wheeler in the TNT miniseries Into the West. From 2007 to 2012, he played Rufus Humphrey, the father of Dan and Jenny, on the CW teen drama series Gossip Girl. Additional screen credits include a guest appearance as a reporter in the seventh-season Law and Order: Special Victims Unit episode "Storm," the role of John in the film Beneath, and the role of John in the 2006 film The Celestine Prophecy.

In 2006, at age 37, Settle eloped with Israeli actress and model Naama Nativ. He publicly disclosed the marriage in 2007. The couple had a child together in 2009 and divorced in May 2011.
