The eldest child of sculptor Don Gummer and actress Meryl Streep, she grew up in Salisbury, Connecticut, and spent a portion of her childhood in Los Angeles alongside her older brother, Henry Wolfe Gummer, and younger sisters, actresses Grace Gummer and Louisa Jacobson. As a toddler, she appeared with her mother in the 1986 film Heartburn, credited under the name Natalie Stern; she was twenty months old during filming and received a positive notice in The New York Times. Gummer attended Miss Porter's School before graduating from the Kent School in Kent, Connecticut, and subsequently studied theater and communications at Northwestern University, earning her degree in 2005.
Following her graduation, Gummer made her off-Broadway debut opposite Michael C. Hall in the premiere of Noah Haidle's Mr. Marmalade, a performance that earned her a Theatre World Award in 2006. She also participated in The 24 Hour Plays in 2004. In 2007, her portrayal in Theresa Rebeck's The Water's Edge brought her a Lucille Lortel Award nomination. Gummer made her Broadway debut in the Tony Award-nominated revival of Les Liaisons Dangereuses, which ran from 2007 to 2008. From October to December 2015, she starred in the original production of Ugly Lies the Bone for the Roundabout Theater Company, and her performance earned her a 2016 Drama Desk Award nomination for Outstanding Actress in a Play.
Her adult film career began with a minor role in Lasse Hallström's The Hoax in 2006. The following year, she appeared in Evening, a film adaptation of Susan Minot's novel directed by Lajos Koltai, in which she played a younger version of her mother's character alongside Vanessa Redgrave, Glenn Close, and Claire Danes. Subsequent film credits include Steven Soderbergh's Side Effects in 2013, Cake in 2014, and Ricki and the Flash in 2015. She starred in the ABC medical drama Off the Map beginning in 2011, and in 2012 took the title role in The CW series Emily Owens, M.D., which completed its 13-episode order before cancellation. Beginning in 2015, she held a featured role in the WGN America series Manhattan. She also appeared in the third season of HBO's anthology drama True Detective, playing Lucy Purcell. In April 2011, she starred alongside Hamish Linklater and Alison Fraser in the Off-Broadway production of The School for Lies, written by David Ives, directed by Walter Bobbie, and produced by Classic Stage Company.

A separation was announced in March 2013. She became engaged to writer Mehar Sethi in August 2018, and the two married in February 2019, the same month their son was born.
