Herbert Jefferson Jr. was born on September 28, 1946, in Sandersville, Georgia, and was raised in Jersey City, New Jersey, where he attended Lincoln High School. He briefly enrolled at Jersey City State College and Rutgers University before earning a degree with honors from the American Academy of Dramatic Arts in New York in 1969. His formal training extended to the HB Studio in New York, where he studied with William Hickey and Michael Becket, and to The Actors Studio under Lee Strasberg.

Jefferson's stage career brought him to Broadway between 1968 and 1971, with credits that include Murderous Angels and The Great White Hope. His feature-film debut came in 1971 with Roger Corman's Private Duty Nurses, in which he played activist Dr. Elton Sanders. He subsequently appeared in two films alongside football legend Jim Brown: Black Gunn in 1972, where he portrayed Brown's brother, and The Slams in 1973, in which he played Brown's cellmate. Later film work included Ron Howard's Apollo 13 in 1995, where Jefferson appeared as a news reporter, and Wolfgang Petersen's Outbreak, also released in 1995, in which he played a doctor.

On television, Jefferson is perhaps most widely recognized for his portrayal of Lieutenant Boomer on the original Battlestar Galactica series. The role came about after the originally intended actor, Terry Carter, was injured in a roller-skating accident; Carter was subsequently cast as Colonel Tigh in the same series. In the 1980 spin-off Galactica 1980, Jefferson's character was promoted to colonel and elevated to the position of executive officer of the Galactica, effectively replacing Carter's character in the chain of command. Jefferson also co-starred as merchant seaman Roy Dwyer in the television novel Rich Man, Poor Man and its sequel, Rich Man, Poor Man Book II. His additional television credits include the Emmy Award-winning miniseries The Law, opposite Judd Hirsch, in which he played Jamaican defendant Maxwell Fall, and The Bastard, a Revolutionary War television film based on John Jakes's novel, where he appeared as Lucas, one of the Minutemen. He guest-starred in the pilot episode of the 1982 series Knight Rider as Muntzy, an undercover police officer and partner of Michael Long, and in the 1984 pilot of Airwolf as the commander of the aircraft carrier USS Enterprise.

Jefferson's work extended into web-based productions as well. In 2007, he portrayed Captain Galt, a freedom fighter, in the independent Internet miniseries Star Trek: Of Gods and Men. In 2015, he appeared as Admiral Grant Satterlee in the pilot of Star Trek: Renegades, a web series whose pilot had originally been developed for television before being made available online.
