Elizabeth Egan Gillies was born on July 26, 1993, in Haworth, New Jersey, to Dave and Lorraine Gillies. She has one younger brother. Gillies has Irish ancestry and an Italian grandmother and great-grandmother, and her paternal grandfather, John Egan Gillies, served in World War II. She attended Northern Valley Regional High School at Demarest for her freshman year before leaving to pursue acting, completing her education through an online program.

Gillies began pursuing acting at age 12 through local open casting calls and soon appeared in commercials for companies including Virgin Mobile. Her first television role was a recurring part across three episodes of The Black Donnellys. In 2008, she appeared in the films The Clique, Harold, and Locker 514, and was cast as Lucy in a Goodspeed production of Jason Robert Brown's musical 13, which also featured Ariana Grande. The production subsequently transferred to Broadway later that year, becoming the first Broadway production to feature a cast and band composed entirely of teenagers. Gillies remained with 13 through its closure on January 4, 2009.

In 2010, Gillies was cast as Jade West in the Nickelodeon series Victorious, a sitcom set at a performing arts high school in Hollywood that premiered on March 27, 2010, and ran for four seasons until February 2, 2013. During that period she contributed to three soundtrack albums — Victorious, Victorious 2.0, and Victorious 3.0 — including the duet "Give It Up" with Grande and "Take a Hint" with Victoria Justice, the latter earning Gillies a platinum certification from the Recording Industry Association of America and a silver certification from the British Phonographic Industry. She also wrote and recorded "You Don't Know Me" for an episode of the series, which appeared on Victorious 3.0. Concurrently, she voiced the character Daphne on the Nickelodeon revival of Winx Club from 2011 to 2014 and recorded the series' official song, "We Are Believix." In December 2013, she recorded a duet of "Santa Baby" with Grande for Grande's Christmas Kisses EP.

Beginning in 2012, Gillies took on guest roles outside of Nickelodeon, with appearances on White Collar and The Exes. That same year she was cast as Courtney in a Manhattan reading of a musical adaptation of Jawbreaker, though the production did not advance to Broadway. In 2014, she appeared in the horror film Animal as Mandy, a production filmed in Manchester, Connecticut, and produced by Drew Barrymore, and in the Lifetime film Killing Daddy as Callie Ross. The following year she was cast as Heather in the comedy film Vacation, released on July 29, 2015, and joined the FX comedy series Sex & Drugs & Rock & Roll as Gigi, the daughter of a former rock star played by the show's creator Denis Leary. The series ran for two seasons before FX canceled it on September 9, 2016, and Gillies performed on two soundtracks released in connection with the show.

In 2017, Gillies was cast as Fallon Carrington in The CW's Dynasty, a reboot of the 1980s series of the same name, a role she held through 2022. During the show's second season she also temporarily portrayed the character Alexis Carrington following Nicollette Sheridan's departure from the role. In 2018, she starred in the dark-comedy thriller Arizona as Kelsey, a film that premiered at the 2018 Sundance Film Festival before receiving both a theatrical and direct-to-video release. In 2019, Gillies made a surprise appearance at Grande's Sweetener World Tour show in Atlanta on November 19, performing "Give It Up" alongside Grande.

During the COVID-19 pandemic in 2020, Gillies collaborated with Seth MacFarlane on eight songs that were released as the extended play Songs from Home in 2021. That same year it was announced she would voice the title character in the animated feature Catwoman: Hunted, released in 2022. In 2023, Gillies and MacFarlane released the Christmas studio album We Wish You the Merriest through Republic Records. In February 2025, she returned to the stage, playing Audrey opposite Milo Manheim as Seymour in the Off-Broadway revival of Little Shop of Horrors at the Westside Theatre, with her run scheduled to conclude in July 2025. Also in 2025, Gillies held a concert residency at Café Carlyle in New York.

Gillies met music producer Michael Corcoran on the set of Victorious. The two began dating in late 2012 and married on August 8, 2020, at a private ceremony in Chesterfield Township, New Jersey. They reside in Atlanta, Georgia.
