James Cullen Landis was born on July 9, 1896, in Nashville, Tennessee, the middle child of Lulan and Margaret (née Cullen) Landis, who had two sons and a daughter. His father worked as a stockbroker to support the family. As a child, Landis harbored ambitions of becoming a railroad engineer, and though that goal did not materialize, his fascination with trains persisted into adulthood, eventually leading him to design and build a steam-powered model train set for himself.

Landis entered the film industry around age 18, roughly the same time his older sister, Margaret Landis, made her own screen debut. He began his career as a director rather than an actor, transitioning to performing only after a lead player broke a leg and it was discovered that the injured actor's costumes fit Landis. That circumstance launched what became a substantial acting career. Over the following fourteen years, he appeared in approximately one hundred films and established himself as one of the more prominent leading men of the silent era.

In 1928, Landis starred in Lights of New York, recognized as the first all-talking motion picture. Despite his role in that landmark production, he privately expressed to a friend that he did not consider himself a song and dance man, even as he acknowledged that talking pictures were well suited to musicals. The following year, he made his Broadway appearance in the 1929 play Chippies, adding stage work to his screen credits.

By 1930, Landis had relocated from Hollywood to Detroit, where he shifted his focus to producing and directing industrial films for automobile companies. In 1937 he joined the directorial staff of the Jam Handy Picture Service, an association that would define the remainder of his professional life.

During World War II, Landis served as a captain in the United States Army Signal Corps, producing training films in the South Pacific. He received two decorations for his service and was promoted to the rank of major before the war ended. In the postwar period, he directed documentaries for the United States State Department, work that brought him to locations around the world. He subsequently returned to industrial filmmaking at the Jam Handy Corporation, where he concluded his career.

Landis died on August 26, 1975, at a nursing home in Bloomfield, Michigan, at the age of 79, three months after the death of his wife, Jane (née Greiner).
