Constantine James Maroulis was born on September 17, 1975, in Brooklyn, New York, to Greek American parents, James Constantine and Constance P. Maroulis. Both his maternal and paternal grandparents emigrated to the United States from Greece in the 1920s. Maroulis was raised in the Greek Orthodox faith and speaks Greek. When he was five years old, his family relocated to Wyckoff, New Jersey, a suburb in Bergen County. His older brother, Athan Maroulis, born in 1964, works as a record producer, and his sister, Anastacia P. Maroulis, born in 1968, is a school principal.

Maroulis developed an interest in music at an early age, influenced by his older brother, and began studying trumpet at Eisenhower Middle School by age eleven. He later sang in high school garage bands, including Lady Rain and Milkbone, before graduating from Ramapo High School in 1993. He went on to earn a Bachelor of Fine Arts in Music Theater from the Boston Conservatory. Following graduation, he trained as an acting apprentice at the Williamstown Theatre Festival in the Berkshires of Western Massachusetts. He also appeared in a number of off-Broadway productions and independent films, and performed the role of Roger Davis in the Broadway international touring company of Rent, a non-equity engagement. He was included in the cast recording of the 2000 musical Suburban Dreams, produced by composer Stephen Thompson, Richard Kenner, and Jesse Merz. Prior to his national breakthrough, Maroulis competed on the dating show Elimidate, worked as an extra on Law and Order: Special Victims Unit, and provided voice-over work for Astro Boy.

In August 2004, acting on a suggestion from his then-girlfriend, Maroulis traveled to Washington, D.C., to audition for the fourth season of the reality television competition American Idol. That season, producers expanded the eligible age range from 16–24 to 16–28, a change from which Maroulis and fellow contestant Bo Bice were among the first to benefit. Both were also notable for competing in the rock idiom. Maroulis was eliminated on April 27, 2005, after performing Nickelback's "How You Remind Me" during the 2000s music round, finishing as the sixth-place finalist. His rendition of Queen's "Bohemian Rhapsody" drew praise from the judges, and at the personal invitation of Queen guitarist Brian May, Maroulis recorded a studio version of the track for Killer Queen: A Tribute to Queen, released on August 9, 2005, on Hollywood Records. He was also featured on the American Idol Season 4: The Showstoppers compilation with "My Funny Valentine."

Before his time on American Idol, Maroulis had become the lead vocalist of Pray for the Soul of Betty in 2003, after winning out over more than 150 auditioners. The band gained national attention during his Idol run. On March 20, 2006, after three years with the group, Maroulis announced his departure due to creative and business differences. He subsequently pursued a solo career, performing shows in Manila and New Jersey in late 2005, with a set list drawing from his theater work in productions including Hedwig and the Angry Inch, Rent, and Jesus Christ Superstar, as well as his American Idol performances. In August 2006, he completed a solo tour of thirteen cities. Maroulis founded his own independent label, Sixth Place Records, a branch of Sixth Place Productions, which released his self-titled album Constantine on August 7, 2007. The twelve-track record debuted at number 75 on the Billboard charts.

Maroulis made his Broadway debut in 2006 and remained active on Broadway through 2013. He starred in Rock of Ages, for which he received a Tony Award nomination for Best Performance by a Leading Actor in a Musical in 2009. He also starred in the title role of Jekyll and Hyde on Broadway, earning a Drama League Award nomination for Distinguished Performance. Additional Broadway credits include The Best Little Whorehouse in Texas and the comedy The Silver Wedding. A production in which he was involved received a Tony Award nomination for Best Revival of a Musical in 2016.

Beyond the stage and recording studio, Maroulis has performed the National Anthem at numerous high-profile events. On July 4, 2009, he sang at the official ceremonies marking the reopening of the Statue of Liberty's crown to the public for the first time since its post-September 11 closure. He also performed the anthem at the 2013 Opening Day festivities for the New York Yankees, at Madison Square Garden in April 2009, and before a 49ers/Bengals football game at Monster Stadium in San Francisco in December 2007, a performance noted by commentator Bryant Gumbel. He sang God Bless America at the 2009 Men's U.S. Open.

In October 2008, Maroulis appeared as a contestant on Don't Forget the Lyrics, resulting in a $25,000 donation to Broadway Cares/Equity Fights AIDS. He has been a regular contributor to the Loukoumi children's book series by author Nick Katsoris, providing the voice of Gus the Bear. In July 2011, he became a spokesman for Disney/Fisher-Price's Rock Star Mickey interactive plush toy and released an updated version of the Kinks' "You Really Got Me" in conjunction with the project. In 2015, he recorded a version of the Lita Ford and Ozzy Osbourne duet "Close My Eyes Forever" with the New York-based band SYKA, which was also released as a music video. That September, Maroulis participated in the Rock in Rio festival in Rio de Janeiro. He also formed MarKolTop Productions with Diego Kolankowsky and Joanna Topetzes.
