Conrad Wayne Ricamora-Jensen was born on February 17, 1979, in Santa Maria, California, and raised primarily in Niceville, Florida. His father, Ron Ricamora, was born in Manila, Philippines, and relocated to the United States at age ten; he is believed to be of Filipino descent and was adopted. Ricamora's mother, Debbie Bolender, works as a social worker and is of German and Irish descent. During his youth, Ricamora was an avid tennis player, a pursuit that earned him a scholarship to Queens University of Charlotte, where he graduated with a degree in psychology in 2001. His introduction to acting came during his undergraduate years, and after graduating he participated in community theater in Charlotte, North Carolina, and Philadelphia before pursuing formal training. He later completed an MFA in acting at the University of Tennessee in 2012.

Ricamora's professional stage career began in earnest with the 2013 Off-Broadway musical Here Lies Love at The Public Theater, where he portrayed Ninoy Aquino across two runs of the production. The role earned him a Theatre World Award and a nomination for the Lucille Lortel Award for Outstanding Lead Actor in a Musical. In 2017, he reprised the role of Ninoy Aquino in a production of Here Lies Love at the Seattle Repertory Theater.

He made his Broadway debut in 2015 in Lincoln Center Theater's revival of Rodgers and Hammerstein's The King and I, directed by Bartlett Sher, playing the role of Lun Tha. The Broadway cast recording of that production received a Grammy Award nomination for Best Musical Theater Album. In 2018, Ricamora starred in the world premiere of David Henry Hwang's Soft Power, playing Xue Xing at the Ahmanson Theatre in Los Angeles; the cast recording of that production also earned him a Grammy Award nomination for Best Musical Theater Album, making him a two-time nominee in that category.

In 2024, Ricamora joined the cast of Oh, Mary!, playing Abraham Lincoln opposite Cole Escola as Mary Todd Lincoln. The production transferred to Broadway in June 2024, and his performance earned him a Tony Award nomination for Best Featured Actor in a Play.

Alongside his stage work, Ricamora became widely recognized for his television role as Oliver Hampton on the ABC legal drama How to Get Away with Murder, which ran from 2014 to 2020. He was initially cast in a recurring capacity, became a guest performer in the second season, and was elevated to series regular beginning in the third season, remaining with the show through its sixth and final season.

Ricamora is gay and has been honored with the Human Rights Campaign's Visibility Award. In July 2023, he announced via Instagram that he had married Peter Wesley Jensen earlier that month.
