Charli Grace D'Amelio was born on May 1, 2004, in Norwalk, Connecticut, to Heidi D'Amelio, a photographer and former model, and Marc D'Amelio, a business owner and former Republican Connecticut Senate candidate. She has an older sister, Dixie, who is also a social media personality. D'Amelio began dancing at age three and trained as a competitive dancer for more than a decade. She previously attended King School, a private institution in Stamford, Connecticut, before transitioning to virtual schooling after her social media career took off.

D'Amelio joined TikTok in May 2019, initially posting a lip-syncing video with a friend. Her visibility grew substantially in July 2019 after she posted a duet with user Move With Joy, and expanded further in October 2019 when she performed the "Renegade" dance to K Camp's song "Lottery." Though TikTok users credited her with creating the dance, a New York Times profile later identified Jalaiah Harmon as its originator; D'Amelio subsequently made a practice of crediting the creators of dances she performed. She joined the collaborative content house The Hype House in November 2019 alongside Dixie, the same month she was invited by Bebe Rexha to perform during Rexha's opening set for the Jonas Brothers. By March 2020, D'Amelio had become the most-followed creator on TikTok, displacing Loren Gray, and was the first user on the platform to reach 50 million followers. She held the top position from March 2020 to June 2022 and, as of 2025, remains the second most-followed person on TikTok with over 156 million followers.

In late 2019, former Sony Music executive Barbara Jones signed D'Amelio to her management company, Outshine Talent. In January 2020, D'Amelio and her family signed with United Talent Agency. That February, she appeared in a Super Bowl commercial for Sabra hummus. In March 2020, she and Dixie partnered with UNICEF for an anti-bullying campaign and appeared in the Nickelodeon special #KidsTogether: The Nickelodeon Town Hall, hosted by Kristen Bell. That same month, she partnered with Procter & Gamble to launch the #DistanceDance challenge on TikTok, aimed at encouraging social distancing during the COVID-19 pandemic; the campaign drew praise from Ohio Governor Mike DeWine and earned D'Amelio a Streamy Award nomination for Social Good Campaign. In April 2020, she appeared in the ABC special The Disney Family Singalong during a performance of "We're All In This Together" from High School Musical.

D'Amelio made her feature film debut voicing the character Tinker in the animated film StarDog and TurboCat, released in the United States in June 2020. That same month, she and Dixie partnered with Morphe Cosmetics to launch the makeup line Morphe 2. In August 2020, the sisters released a nail polish collection called Coastal Craze with Orosa Beauty. Dunkin' Donuts introduced a limited-time menu item called "The Charli" in September 2020, based on her regular order. That October, she appeared in the music video for Bebe Rexha's single "Baby I'm Jealous" and received three nominations at the 10th Streamy Awards, including Creator of the Year. In November 2020, she became the first TikTok user to surpass 100 million followers and earned a nomination for Social Star at the 46th People's Choice Awards. She won the Breakout Creator award at the 10th Streamy Awards in December 2020.

In May 2021, D'Amelio and Dixie co-created Social Tourist, an apparel brand developed in partnership with Hollister and Abercrombie & Fitch, and collaborated with Simmons Bedding Company on a mattress release. D'Amelio starred alongside her family in the Hulu docuseries The D'Amelio Show, which premiered in September 2021 and ran for three seasons before being cancelled in June 2024. The series won an MTV Movie & TV Award for Best Unscripted Series. She also co-led the Snap Original reality show Charli vs. Dixie with her sister from 2021 to 2022. In 2022, D'Amelio won the thirty-first season of Dancing with the Stars alongside partner Mark Ballas. Forbes named her the highest-earning personality on TikTok in both 2019, among female creators, and again across all creators in 2022.

D'Amelio's debut book, Essentially Charli: The Ultimate Guide to Keeping It Real, was published by Abrams Books on December 1, 2020, and became a New York Times bestseller. Her additional ventures include a podcast announced in May 2020 through a deal with Ramble Podcast Network, a bracelet range with Pura Vida Bracelets released in March 2021, and a multi-product company encompassing a clothing line and other consumer products.

On the stage, D'Amelio made her Broadway debut in October 2024, joining the ensemble of the musical & Juliet. The production marked her first appearance on Broadway, adding a live performance credit to a career that had previously spanned social media, television, film, and competition. D'Amelio is originally from Norwalk, Connecticut.
