Betty Lynn Buckley was born on July 3, 1947, in Big Spring, Texas. At age 11, a performance of The Pajama Game at Fort Worth's Casa Mañana set her on a path toward musical theater. Her mother, a former tap dancer, quietly enrolled her in dance lessons without the knowledge of her father, who opposed a career in show business. Buckley attended Arlington Heights High School, graduating at 16, and enrolled at Texas Christian University at 17. Her first professional stage role came at 15, when she played Dainty June in a production of Gypsy. In 1966, she won the Miss Fort Worth title and placed as runner-up in the Miss Texas pageant, a performance on the Miss America national telecast that attracted the attention of talent agents who signed her before she completed her senior year. She worked as a reporter at the Fort Worth Press from 1967 to 1968 and earned a Bachelor of Arts in journalism from TCU in 1968, after which she joined a USO tour to South Korea and Japan.

Buckley made her Broadway debut in 1969 in the original production of 1776, launching a stage career that would span nearly five decades. She returned to Broadway in 1973 with Pippin, and in 1985 appeared in The Mystery of Edwin Drood. Her most celebrated Broadway achievement came in 1982, when she originated the role of Grizabella in the American premiere of Andrew Lloyd Webber's Cats, performing the signature song "Memory" and remaining with the production for 18 months. That performance earned her the 1983 Tony Award for Best Featured Actress in a Musical. She later appeared in the Broadway production Andrew Lloyd Webber — Music of the Night, and in December 2006 sang "Memory" at the Kennedy Center Honors as part of the tribute to Lloyd Webber.

In 1988, Buckley starred in the short-lived Broadway musical adaptation of Carrie, playing Margaret White, the mother of the title character. That same year, she had previously played the role of Miss Collins, Carrie's gym teacher, in the 1976 film version. Also in 1988, she appeared in both Roman Polanski's Frantic and Woody Allen's Another Woman. Her 1983 film work included Bruce Beresford's Tender Mercies, in which she played country music singer Dixie Scott and performed the Academy Award-nominated song "Over You."

Buckley starred as Norma Desmond in Sunset Boulevard in London in 1994 and in New York in 1995, receiving a 1995 Olivier Award nomination for Best Actress in a Musical for her work in the London production. In 1997, she returned to Broadway in Triumph of Love, playing Hesione, a role that earned her a Tony Award nomination for Best Actress in a Musical. Over the course of her career, Buckley has accumulated nominations for two Daytime Emmy Awards, two Grammy Awards, and an Olivier Award, in addition to her Tony win. On January 28, 2013, she was inducted into the American Theater Hall of Fame.

Beyond her stage work, Buckley is widely recognized for her role as Sandra Sue "Abby" Abbott on the television series Eight Is Enough, which ran from 1977 to 1981. She joined the show in its second season after the original star, Diana Hyland, died following the first four episodes. Her character became the romantic interest and eventual stepmother figure to the eight children central to the series. From 2001 to 2003, she played Suzanne Fitzgerald in seasons four through six of the HBO series Oz, and she appeared in season three of Preacher in 2018. Her television work also includes a role in the HBO miniseries The Pacific in 2010, guest appearances on Without a Trace, Law and Order: Special Victims Unit, Monk, and The Leftovers, and five episodes of Supergirl between 2017 and 2021. She received two Daytime Emmy Award nominations for Outstanding Performer in a Children's Program or Special, for NBC Special Treat — Bobby and Sarah in 1984 and ABC Afterschool Special — Taking a Stand in 1989.

On film, Buckley played Mrs. Jones in M. Night Shyamalan's The Happening in 2008 and Dr. Karen Fletcher in Shyamalan's Split in 2016, the latter earning her a Saturn Award nomination. Additional film credits include Dixie Scott in Tender Mercies, Sondra Walker in Frantic, Kathy in Another Woman, and a role in Lawrence Kasdan's Wyatt Earp in 1994.

In September 2013, Buckley starred in the world premiere of Horton Foote's The Old Friends at the Signature Theatre off-Broadway, reprising the performance in 2014 at Houston's Alley Theatre. She played Big Edie in Grey Gardens at Bay Street Theater in 2015 and reprised that role at the Ahmanson Theatre in Los Angeles in 2016. From October 2018 through September 2019, she starred as the title character in the first national tour of the 2017 Broadway revival of Hello, Dolly. Buckley is also a recording artist, with Hope, her 18th solo album, released on June 8, 2018, and she continues to perform in concert across the United States.
