Thomas Arthur Darvill was born on 17 June 1982 in Birmingham, England. His mother, Ellie, worked as an actress with Cannon Hill Theatre at the Midlands Arts Centre, performing with masks, puppets, and live acting, and is also recognized as the puppeteer and voice behind Why Bird from the children's television program Playdays. His father, Nigel, was a Hammond organ player who performed with artists including Edwin Starr, Ruby Turner, Fine Young Cannibals, and UB40. Darvill attended Bromsgrove School in Worcestershire from 1993 to 2000 and joined Stage2 Youth Theatre Company at age ten, remaining a member until 2000. He later trained in acting at the Royal Academy of Dramatic Art, where he also received training in stage combat.

Darvill made his professional stage debut in Edmund White's Terre Haute, playing condemned criminal Harrison at the Assembly Rooms during the Edinburgh Festival Fringe. The production transferred to Trafalgar Studios in 2007, and his performance earned him a Best Newcomer nomination at the 2007 Evening Standard Theatre Awards. That same year he played Rob in the monologue Stacy and appeared at the Vaudeville Theatre in Swimming with Sharks alongside Christian Slater, Helen Baxendale, and Matt Smith, a role that brought him a London Newcomer Award nomination.

His television career began in 2008 with the ITV crime drama He Kills Coppers, followed by the role of Edward "Tip" Dorrit in the BBC serial Little Dorrit. Darvill went on to portray Rory Williams, companion to the Eleventh Doctor, beginning in the fifth series of Doctor Who, reuniting him with his Swimming with Sharks co-star Matt Smith. He remained a regular through the seventh series before departing alongside co-star Karen Gillan. Following Doctor Who, he was cast as Reverend Paul Coates in the drama Broadchurch, appearing opposite David Tennant and Olivia Colman from 2013 to 2017. In March 2015, he was cast as time-traveller Rip Hunter in Legends of Tomorrow, a role he held through the end of the third season in 2018 before returning for the show's hundredth episode in 2021.

On Broadway, Darvill took over the lead male role of Guy in the musical Once on 19 April 2013, performing opposite Joanna Christie. He and Christie also appeared at the 2013 Tony Awards, performing a number and presenting the award for best scenery. Darvill subsequently continued the role in the West End production of Once from 17 March 2014 to 10 May 2014. In 2019, he played Oscar Lindquist in the West End revival of Sweet Charity. In 2022, Darvill took on the role of Curly McLain in Oklahoma! at the Young Vic, continuing the role in the West End production in 2023 and winning the Laurence Olivier Award for Best Actor in a Musical for his performance.

Beyond acting, Darvill works as a composer and musician. He wrote songs and music for the Bush Theatre and composed the score for Che Walker's 2008 play The Frontline at Shakespeare's Globe. His collaboration with Walker on the musical The Lightning Child, which opened at the Globe in September 2013, earned him a Judge's Discretionary Award from MTM. In 2016, he collaborated with playwright Sam Holcroft on a musical adaptation of Roald Dahl's Fantastic Mr. Fox, which premiered at the Nuffield Theatre in Southampton. In 2018, he co-founded the musical collective Paperlands with Inès De Clercq, with whom he performed at St Pancras Old Church in November of that year. Darvill is also set to portray Beatles press officer Derek Taylor in The Beatles – A Four-Film Cinematic Event, a series of biopics directed by Sam Mendes.
